Target Information

Thymidine kinase 1 (TK1) is a crucial enzyme involved in the DNA synthesis pathway, specifically catalyzing the phosphorylation of thymidine to thymidine monophosphate (dTMP), which is further phosphorylated to thymidine triphosphate (dTTP). TK1 is located on chromosome 17q25.3 and is expressed predominantly during the S-phase of the cell cycle. Structurally, TK1 can form tetramers, which enhances its enzymatic activity. Functionally, TK1 is upregulated in various cancers, making it a potential biomarker for tumor diagnosis and prognosis. It plays a significant role in DNA repair, especially in p53-deficient tumor cells, by expanding the dTTP pool during recovery from DNA damage. Additionally, TK1 influences oncogenic phenotypes by promoting glycolysis through interactions with protein arginine methyltransferase 1 (PRMT1). Targeting TK1 expression is considered a promising therapeutic approach to enhance anti-tumor immunotherapy efficacy.
